CRISPR-Cas system has been widely utilized into genome engineering and many scientists have struggled to apply it especially for human beings. However, there are critical limitations in it. Those are Specificity and Activity. Specificity should be increased even up to “100%” not to make patients suffering from side effects and from death. Activity should be improved for efficient preparation of desired cells and also the activity should be properly controlled to reduce the “off-target effects”. We approach the issues by engineering and designing the nucleic acids regions of CRISPR-Cas system such as sgRNA (single guide RNA).
